Abstract：Objective To investigate the effect andmechanism of Huoxin pill(concentrated pill)onatherosclerosis ( As) in apolipoprotein E knockout ( ApoE-) mice. Methods ApoE" mice were fed withhigh-fat diet to replicate As model. "The miee were treated with different concentrations of Huoxin pills andToll-like receptor 4(TLR4)inhibitor. The micewere randomly divided into control group, Huoxin piltreatment group, TLR4 inhibitor treatment group .and Huoxin pill and TLR4 inhibitor co-treatment group,Hematoxylin-eosin(HE) staining and Masson staining were used to observe the pathological changes olaortic sinus of As model mice, Oil red 0(0RO) staining was used to observe the lipid accumulation inaortic sinus and aorta of As model mice. lmmunohistochemistry was used to detect the expression of TLR4and tumor necrosis factor-0 ('TNF-0r ) in aortic sinus of As model mice. Results Compared with the controlgroup, Huoxin pills significantly reduced the aortic sinus lesions, reduced the lipid accumulation in theaortic lesions and the degree of aortic sinus stenosis of As model mice, with a statistically signilicantdifferenee ( P<0.05 ). In addition, Huoxin pills also down-regulated the expression of TLR4 and TNF-a inaortic sinus of As model mice( P<0.05 ), and the efeet of TLR4 inhibitor and Huoxin pills had asynergistic effeet. Conclusion Huoxin pills can alleviate As in ApoE" mice and its mechanism may berelated to down-regulating the expression of 'TLR4 and 'TNF-o and regulating the inflammatory response.Keywords: Huoxin pill; atherosclerosis; Toll like receptor 4; tumor necrosis factor-a
